Bangkok: Faculty of Tropical Medicine, Mahidol University; 2005. p. 168.Cholroquine resistance of Plasmodium falciparum is associated with the mutation in a transporter gene found on food vacuole membrane (chloroquine resistance transporter, CRT). PfcrtT76 mutation is strongly correlated with in vitro cholroquine resistance. In this study, PfcrtT76 mutation was investigated from 130 blood samples from falciparum malaria patients sdmitted at the Hospital for Tropical Diseases, Faculty of Tropical Medicine, Mahidol University, Bangkok, Thailand by real-time PCR assay using LightCycler. Parasite DNA of each blood sample was extracted and purified by High Pure PCR Template Preparation Kit (Roche Applied Science). Pfcrt T76 mutation was detected by using Pfcrt specific primers labeled with LCRed 640 and fluorescent-labeled hybridization probes. T76 Pfcrt mutation was detected by melting temperature curve analysis and an assay can be completed in an hour. In our study, Pfcrt T76 mutation was found in all specimens. The result shows an existence of the high prevalence of chloroquine resistance in this area. Moreover, a real-time PCR can demonstrate mixed infections of wild-type and mutant P.falciparum strains. Based on the results, real-time PCR LightCycler-based technique is rapid, informative and useful for molecular surveillance of chloquine resistance of P.falciparumengMahidol UniversityMalariaPFCRT T76Detection of PFCRT T76 Mutation in falciparum Malaria patients from Thailand by real-time PCR ASSAYProceeding Book
