There is no requirement in Pharmacopoeia for dissolution testing of ketoconazole tablets whrch is a wrdely used as antifungal in Thailand. Thus, it is essential to establish a simple method for dissolution testing to ensure that a generic drug at low cost is at least dissolute as readily as the original brand. An optimum UV wavelength that is the maximally absorbed by a standard solution of ketoconazole appears to be 268 nm. The USP dissolution apparatus ll rotating at Z5 rpm and 0.1 M HCI maintained at 37oC were used as the dissolution medium to determine the dissolution profile of the ketconazole tablets (original brand). The results demonstrate that the above described method is reliable and can be used to determine the dissolution rate of ketoconazole tablet.อัตราการละลายของตัวยาจากยาเม็ดเป็นปัจจัยสำคัญในการที่จะบ่งบอกถึงอัตราและ/หรือปริมาณการดูดซึมของยานั้นเข้ากระแสโลหิต แต่ไม่มีข้อกำหนดนี้สำหรับยาเม็ด ketoconazole ในตำรายาต่าง ๆ คณะผู้วิจัยจึงพัฒนาวิธีที่สะดวกและง่ายต่อการทดสอบหาอัตนาการละลายของตัวยาจากยาเม็ด ketoconazole โดยใช้ spectrophotometer วัดการดูดกลืนรังสีอุลตราไวโอเลตที่ความยาวคลื่น 268 นาโนเมตร และใช้เครื่องทดสอบอัตราการละลายตามที่บ่งไว้ใน USP XXIII แบบ Apparatus II โดยใบพัดหมุนด้วยอัตรา 75 รอบต่อนาที และใช้ 0.1 M HCI เป็นสารละลาย ควบคุมอุณหภูมิที่ 37 องศาเซลเซียส ผลที่วัดได้ผลการวิเคราะห์ว่าวิธีที่กล่าวข้างต้น สามารถใช้วัดอัตราการละลายของตัวยาจากยาเม็ด ketoconazole ได้ และผลการทดสอบการละลายตัวยา ketoconazole ที่ละลายจากยาเม็ดที่ผลิตโดยบริษัทต้นแบบ พบว่าตัวยาละลายได้ 96.54 เปอร์เซ็นต์ที่เวลา 15 นาที243 KBapplication/pdfengMahidol UniversityKetoconazoleDisolutionRate-limiting stepResearch ArticleFaculty of Pharmacy Mahidol University.
