จิตติมา โชติวรานนท์บุษบา ฤกษ์อำนวยโชคJittima ShotivaranonBudsaba Rerkamnuaychokeมหาวิทยาลัยมหิดล. คณะแพทยศาสตร์โรงพยาบาลรามาธิบดี. ภาควิชาพยาธิวิทยา2022-07-212022-07-212565-07-212565รามาธิบดีเวชสาร. ปีที่ 45, ฉบับที่ 1 (ม.ค.-มี.ค. 2565), 43-570125-3611 (Print)2651-0561 (Online)https://repository.li.mahidol.ac.th/handle/123456789/72199การตรวจพิสูจน์เอกลักษณ์บุคคลสามารถนำมาประยุกต์ใช้ในการพิสูจน์ความสัมพันธ์ระหว่างบุคคล โดยใช้ชุดน้ำยาสำเร็จรูปซึ่งผลิตบนหลักการเพิ่มปริมาณดีเอ็นเอหลายตำแหน่งพร้อมกันในหลอดทดลองหนึ่งหลอดที่ตรวจวิเคราะห์สารพันธุกรรมลำดับซ้ำ (Short tandem repeat, STR) ชุดน้ำยาสำเร็จรูปแต่ละชนิดมีความจำเพาะและข้อจำกัดที่แตกต่างกัน การศึกษานี้มีวัตถุประสงค์เพื่อเปรียบเทียบจุดเด่นและข้อจำกัดของชุดน้ำยาสำเร็จรูปที่นิยมใช้ในประเทศไทย จำนวน 8 ชุดน้ำยา (GlobalFiler Express, GlobalFiler™ IQC/GlobalFiler, Verifiler Express, Verifiler Plus, Investigator 24 Plex QS, Investigator 24 Plex Go, Investigator 26 Plex QS และ PowerPlex Fusion6C) รายงานนี้นำเสนอตัวอย่างการใช้งานของชุดน้ำยาสำเร็จรูปจำนวน 3 ชุดน้ำยา จากงานบริการพิสูจน์ความสัมพันธ์บิดา-มารดา-บุตร ของห้องปฏิบัติการมนุษย์พันธุศาสตร์ ภาควิชาพยาธิวิทยา คณะแพทยศาสตร์โรงพยาบาลรามาธิบดี มหาวิทยาลัยมหิดล พบว่าการใช้ชุดน้ำยาสำเร็จรูปที่แตกต่างกันมากกว่า 1 ชุดน้ำยา สามารถแก้ปัญหาการแปลผลการตรวจวิเคราะห์ที่มีการกลายพันธุ์หรือพบข้อขัดแย้งได้ในบางสถานการณ์Human identification is applied to prove interpersonal relationships by using commercial test kits of a single tube multiplex PCR on DNA repeated sequence (short tandem repeat, STR) genetic markers. Each commercial test kit has different limitation and advantage. The objective of this report is to compare the advantage and limitation of 8 commercial test kits (GlobalFiler Express, GlobalFiler™ IQC / GlobalFiler, Verifiler Express, Verifiler Plus, Investigator 24 Plex QS, Investigator 24 Plex Go, Investigator 26 Plex QS, and PowerPlex Fusion6C). We not only reviewed and compared the strengths and limitations of these 8 commercial test kits but also compared DNA profiles generated by 3 commercial test kits which were used in routine paternity service of Human Genetic Laboratory, Department of Pathology, Faculty of Medicine Ramathibodi Hospital, Mahidol University.
