Mahidol University Library and Knowledge Center, 2024) Nipa Walaiphachara; Kana Romruen; Punya Temcharoen; Aimon SomanabandhuThe study of effect of the ethanolic extract of Phyllanthus amarus on carbon tetrachloride (CC1(,4)) induced hepatotoxicity in Spraque-Dawley male rats was carried out.The animals were given the extract of Phyllanthus amarus before administration of carbon tetrachloride. In this study, we investigated the effect of Phyllanthus amarus on growth, nutritional status and some possible toxicities in rats after administration of various doses of Phyllanthus amarus (5.0,1.0,andO.2 ml/kg bw.)for 28 days. The results revealed that rats receiving Phyllanthus amarus 5.0 ml/kg bw had a decrease in growth rate and food intake in comparison to the control group. The levels of AP,BUN, total protein and plasma glucose were significantly higher than the control group (P<0.05). However, there was no hematologic changes in rats receiving Phyllanthus amarus 5.0 ml/kg bw.These evidence indicated that the administration of Phyllanthus amarus 5.0 ml/kg bw.for 28 days had some toxic effects on rats. The inhibition of Phyllanthus amarus on hepatotoxicity induced by carbon tetrachloride was observed when Phyllanthus amarus (1.0 ml/kg bw.)was administered for 28 days before carbon tetrachloride(2.5 ml/kg bw.)was given.It was found that Phyllanthus amarus could reduced hepatotoxicity by lowering liver enzymes(GOT and GPT) when compared with the control group given carbon tetrachloride alone (P<0.05). In view of histopathologic study, it was revealed that administration of Phyllanthus amarus (1.0 ml /kg bw.)decreased the necrotic hepatocytes from the toxic effect of carbon tetrachloride.On the other hand, in the group given Phyllanthus amarus (5.0 ml/kg bw.) and carbon tetrachloride, it appeared that Phyllanthus amarus induced more hepatotoxicity than the control group. The amount of 2 in the herb juice was quantitatively determined by use of a validated HPLC method (RP-18, MeOH-H2O, UV detection at 210 nm). The immunomodulatory effect of the herb juice and of 2 was proved by means of in vitro cellular immunological assays. Phytochemicals in herbal medicine may interact with cytochromes P450 (CYP) and thus raise the potential of herb-drug interactions; therefore, the inhibitory effects of P. amarus and its major phytochemicals phyllanthin and hypophyllanthin on CYP isoforms were determined using human liver microsomes and selective substrates. Both ethanolic and aqueous extracts of P. amarus inhibited CYP1A2, CYP2D6, CYP2E1 and CYP3A4 in a dose-dependent manner. Compared to known CYP3A inhibitors, the IC 50 values of the ethanolic and aqueous extracts on testosterone 6β-hydroxylation were higher than that of ketoconazole but were lower than those of erythromycin and clarithromycin. Both extracts were weak inhibitors of CYP1A2, CYP2D6 and CYP2E1. In addition, phyllanthin and hypophyllanthin were potent mechanism-based inhibitors of CYP3A4 with K I values of 1.75 ± 1.20 μM and 2.24 ± 1.84 μM and kinact values of 0.18 ± 0.05 min -1 and 0.15 ± 0.06 min -1 , respectively. The k inact /K I ratios of these lignans were higher than those reported for some therapeutic drugs that act as mechanism-based inhibitors of CYP3A4. Five species of Cyphomandra examined all contained both amines and amides. From roots of Margaranthus solanaceus cuscohygrine has been isolated which probably occurs, too, in roots of Lycianthes rantonnettii. Cordell; Mahidol University; University of Illinois at ChicagoComplete 1H-nmr data and unambiguous assignments of the 13C-nmr spectra of phyllanthin [1] and hypophyllanthin [2] were obtained through extensive nmr studies, including homonuclear COSY, homonuclear decoupling, APT, HETCOR, nOe difference, selective INEPT, and COLOC experiments. The absolute configuration of hypophyllanthin [2] was determined by cd. Neither of these lignans demonstrated significant cytotoxic activity when evaluated with a battery of cultured mammalian cells, but both were found to enhance the cytotoxic response mediated by vinblastine with multidrug-resistant KB cells. In addition, 1 was found to displace the binding of vinblastine with membrane vesicles derived from this cell line, suggesting an interaction with the P-glycoprotein. © 1993, American Chemical Society.
