Protein and lipid composition of sarcoplasmic reticulum from dystrophic muscles of vitamin E-deficient rabbits
The protein and lipid compositions of sarcoplasmic reticulum (SR) membranes from vitamin E-deficient and control rabbits were analyzed. In vitamin E-deficient rabbits, sodium dodecyl-sulfate polyacrylamide gel-electrophoresis and sulfhydryl content of SR proteins and lipid composition were normal, except that cholesterol and sphingomyelin levels were increased. SR calcium-dependent ATPase (EC 3.6.1.3) activity from vitamin E-deficient rabbit was reduced compared with control. Elevating the cholesterol content of control SR membranes by incubating with cholesterol-enriched egg lecithin liposomes to a levels comparable to that of vitamin E-deficient SR membranes produced a corresponding decrease in Ca-ATPase activity.
