Abstract
Sequence analysis of flagellin structural genes from different genera of bacteria demonstrated that the terminal regions are conserved. The central part has been suggested to be hypervariable in sequence, contributing to antigenic diversity. In this report, we attemp to study the divergence of fiagellin gene of Pseudomonas putida from different clinical isolates. Primers designed from the N- and C-terminal conserved regions of 2 kb the flagellin A of P. putida PaW8 were used to amplify the flagellin structural genes of 4 clinical isolates. Polymerase chain reaction amplification showed the three distinct DNA fragments of 2 kb (from 1 isolate), 1.4 kb (from 1 isolate) and 0.7 kb (from 2 isolates). Sequencing analysis revealed tha N- and C-terminal sequences are conserved for flagellin protein. The diversed sequences were found in the central part suggesting to be given rise to serotype-specific-epitopes or contributed to antigenic diversity.
