Abstract
© 2017, International Society for Horticultural Science. All rights reserved. An in vitro plant regeneration protocol was successfully established for Cymbidium finlaysonianum Lindl., an epiphytic orchid by culturing seeds from six-month-old green self-pollinated fruits. Tetrazolium (TZ) viability test indicated a mean embryo viability of 97.80%. Protocorm-like bodies (PLBs) of Cymbidium finlaysonianum Lindl. can be induced from protocorm segments cultured in VW liquid medium supplemented with 8.84 µM BAP and significantly different from other treatments, with the highest number of 5.2 PLBs explant-1. The PLBs converted into plants with well-developed shoots and roots on VW medium supplemented with 0.2% (w/v) activated charcoal (AC), without plant growth regulators after 2 months of culture. The highest percentage of plantlet survival recorded at 2 months after hardening in the greenhouse was 85.8%.
