Abstract
Blood vessel tissue engineering has arisen to serve the problem of vascular diseases. The blood vessel tissue engineering process requires stem cell technology for autologous transplantation. There are many sources of stem cells in adult humans but the most popular are bone marrow stem cells (BMSCs) or mesenchymal stem cells. In this study, for further tissue engineering application, the BMSCs have been induced by TGF-β and/or retinoic acid (RA) for differentiation into smooth muscle cells. The smooth muscle alpha actin (SM α-actin), the earliest smooth muscle cell marker, was observed in induced BMSC in all treatments. The TGF-β1 treatment showed morphological change appearing as a ribbon shape with intracellular stress fiber. The RA treatment showed spindle shaped cells with a tendency to cluster which is similar to the combination and serial addition of TGF-β1 and RA. The expression of SM α- actin was demonstrated using immunofluorescent technique and Western blot analysis. Using TGF-β1 and 0.01μM RA a highly positive reaction for SM α-actin was shown in immunofluorescent analysis. The expression level of SM α-actin was evaluated by Western blot and densitometric analysis. It was shown that TGF-β1 can promote a high expression of SM α-actin. The RA showed good enhanced protein expression at 0.01 μM dose. The BMSCs treated with TGF-β1 for 24hrs and then followed by RA treatment on day 3 promoted expression of SM α-actin at higher degrees than other treatments but on day 5 this protein became lower. The combination at TGF-β1 and RA showed the lowest expression of SM α-actin. This data shows that both TGF-β1 and 0.01μM RA can promote SMC characteristics from BMSC. The study shows that TGF-β1 and RA may regulate SM α-actin expression differently. The BMSCs have shown to be a good source for SMC differentiation. The retinoic acid is another factor which can promote this step.
เทคนิควิศวกรรมเนื้อเยื่อหลอดเลือดและเทคโนโลยีเซลล์ต้นกำเนิดได้ถูกพัฒนาขึ้น เพื่อที่จะใช้ในการปลูกถ่ายให้กับผู้ป่วยที่มีปัญหาความบกพร่องของหลอดเลือด มีเซลล์ต้นกำเนิด หลายชนิดที่สามารถพบได้ในวัยเจริญพันธ์แต่ที่นิยมศึกษามากคือ เซลล์มีเซนครายจากไขกระดูก ซึ่ง ได้นำมาใช้ในการศึกษาครั้งนี้เพื่อส่งเสริมลักษณะของเซลล์กล้ามเนื้อเรียบซึ่งเป็นองค์ประกอบสำ คัญของหลอดเลือดภายใต้การเติม TGF-β1 และ หรือ RA และประเมินลักษณะของเซลล์กล้ามเนื้อ เรียบด้วยการแสดงออกของ SM α-actin ซึ่งเป็นลักษณะของเซลล์กล้ามเนื้อเรียบระยะเริ่มต้น พบว่า เมื่อเลี้ยงเซลล์ต้นกำเนิดจากไขกระดูกด้วย TGF-β1 เซลล์มีการเปลี่ยนแปลงรูปร่างเป็นแบบ คล้ายริบบิ้นและมี stress fiber ภายใน ขณะที่กลุ่มของ RA จะมีการเปลี่ยนแปลงรูปร่างเป็นแบบ กระสวยและมีการจับกลุ่มกันของเซลล์ ซึ่งลักษณะนี้สามารถพบได้ในกลุ่มที่ใส่ TGF-β1 รวมกับ RA และกลุ่มที่เติม TGF-β1 24 hrs ตามด้วย RA. การแสดงออกของ SM α-actin ได้ถูก ประเมินทั้งจากเทคนิค immunofluorescent and Western blot analysis พบว่าจากการเติม TGF-β1 และ 0.01μM RA สามารถเพิ่มการแสดงออกของ SM α-actin เมื่อศึกษาด้วย immunofluorescent ระดับของ SM α-actin จากแต่ละกลุ่มได้ถูกประเมินโดย Western blot and densitometric analysis พบว่า การเติม TGF-β1 สามารถเพิ่มการแสดงออกของ SM α- actin เช่นเดียวกับการเติม RA ในความเข้มขัน 0.01 μM ขณะที่การรวมกันของ TGF-β1 และ RA กลับลดการแสดงออกของ SM α-actin ต่างจากกลุ่ม TGF-β1 24 hrs ตามด้วย RAสามารถ ส่งเสริมการแสดงออกของ SM α-actinได้มากกว่ากลุ่มอื่นในวันที่ 3 และ ลดลงในวันที่ 5 ซึ่ง สามารถบ่งบอกได้ว่า ทั้งสองปัจจัยอาจจะควบคุมการแสดงออกของ SM α-actin แตกต่างกัน จาก การศึกษาครั้งนี้พบว่าเซลล์ต้นกำเนิดจากไขกระดูกเป็นเซลล์ที่ดีสำหรับการส่งเสริมการสร้างเซลล์ กล้ามเนื้อเรียบและ RA เป็นอีกปัจจัยหนึ่งซึ่งสามารถส่งเสริมลักษณะของเซลล์กล้ามเนื้อเรียบจาก เซลล์ชนิดนี้ได้
