Proteomic analysis of differently expressed proteins in penaeus vannamei survivors after acute yellow head virus infection

dc.description.abstractYellow head virus is a highly virulent shrimp pathogen that can cause mass mortalities in farmed penaeid shrimp. To define the change of the protein expression profile of YHV infection, a comparative analysis of proteomic profiles of control and YHV-infected was performed in gill of shrimp by 2-DE. In this proteomic analysis of gills from YHV-infected Penaeus vannamei, we identified 13 spots with up-regulated protein expression levels and 5 spots with down-regulated levels. LC-nanoESI-MS/MS indicated that the up-regulated proteins included enzymes in the glycolytic pathway, the TCA cycle and amino acid metabolism. The other up-regulated proteins were arginine kinase, imaginal disc growth factor (IDGF) and a Ras-like GTP binding protein. By contrast, expression levels were reduced for sarcoplasmic calcimn binding proteins (SCPs), actin-1, Valosin-containing protein and Rabl l. The expression of SCP and enolase by Western blot and irnuno-fluorescence also revealed the change in the level of these proteins after YHV infection corresponding to 2-DE results. However, time-course assays by Real time RT-PCR revealed no significant increase in mRNA level of glycolytic enzymes and arginine kinase except a significant decrease in SCP mRNA. When screening of SCP promoter from fosmid DNA library of P. monodon, some clones from the library possibly contained SCP promoter. The results presented here provide preliminary data on the interaction between YHV and more insights into the pathogenesis and mechanism of YHV infection in P. vannamei in the future.
