Abstract
Fascioliasis gigantica is a parasitic disease in cattle that, in Thailand alone, causes economic damage estimated at approximately 350-400 million bahts per year. The present diagnostic methods are based on the finding and counting of eggs in stool under a microscope, which is unreliable, cumbersome and need well-trained personnel. The aim is to develop immuno-diagnostic assays using specific monoclonal antibodies (MAB) against circulating parasite antigens. In order to reach that goal, the following basic studies have been carried out: ( 1 ) the parasites antigens have been identified, as those originated from the tegument at molecular weights (MW) 66, 58, 54 kD, and those from the caecal epithelium at MW 27,26 kD. The latter have been characterized for their amino acid compositions and sequences, and were identified to be cysteine proteases. (2) The two tissue sources of antigens have been studied by light and electron microscopy as well as immuno-microscopic detection methods, in order to understand the mechanism of antigens syntheses, their distribution and relative amount in the tissues. (3) Monoclonal antibodies (MAB) against excretory/secretory antigens at 58, 54, 27, 26 kD have been produced and their tissue sources identified. The antigens at 58, 54 kD are membrane-associated antigens that are highly concentrated in the surface membrane of the tegument, while lightly distributed in membranes of caecal epithelial and excretory epithelial cells. The antigens at 27 and 26 kD are found in the caecal lumen and the apical region of caecal epithelial cells, thus they are probably digestive enzymes of the parasites. These data were to be used as basis for the selection and testing of these monoclonal antibodies for their potential for devising the immuno-diagnostic methods.
โรคพยาธิใบไม้ตับ Fasciola gigantica เป็นโรคปรสิต ในโคและกระบือที่ทำให้เกิดความเสียหายเป็นมูลค่าประมาณ 350 ถึง 400 ล้านบาทต่อปี ปัจจุบันการตรวจการติดเชื้อทำได้โดย การหาและการนับไข่พยาธิในอุจจาระของสัตว์โดยใช้กล้อง จุลทรรรศน์ ซึ่งเป็นวิธีการที่ไม่สะดวก กินเวลา ไม่แม่นยำ และต้องการบุคคลากรที่ต้องได้รับการฝึกหัดเป็นพิเศษในการ แยกไข่พยาธิ งานวิจัยจึงมุ่งที่จะพัฒนาวิธีตรวจสอบการติดเชื้อ ปรสิตแบบอิมมูโนด้วยการใช้โมโนโคลนัลแอนติบอดีต่อแอนติเจน ที่พยาธิปล่อยออกมาสู่เลือดของสัตว์ที่ติดเชื้อ การพัฒนาวิธี ตรวจสอบได้กระทำเป็นขั้นตอนคือ (1) วิเคราะห์และศึกษา คุณลักษณะของแอนติเจนซึ่งได้แก่ แอนติเจนจากชั้นผิวที่ ประกอบด้วยแอนติเจนหลักมีน้ำหนักโมเลกุล (MW) ที่ 66, 58 และ 54 kD และแอนติเจนจากลำไส้ที่ MW 27 และ 26 kD (2) ศึกษาโครงสร้างของเนื้อเยื่อหลักในพยาธิที่เป็นแหล่ง สร้างของแอนติเจนดังกล่าวซึ่งได้แก่ชั้นผิวและเยื่อบุผิวลำไส้ ของพยาธิด้วยวิธีจุลทรรศน์ธรรมดา จุลทรรศน์อิเลคตรอน และวิธีทางอิมมูโนไซโตเคมีทำให้เข้าใจถึงปริมาณสัมพัทธ์ การกระจายตัวและกลไกการสร้างแอนติเจนในระดับเซลล์ ของแอนติเจนในเนื้อเยื่อเหล่านี้ได้ (3) ผลิตโมโนโคลนัล แอนติบอดีโดยวิธีไฮบริโดมาด้วย excretory/secretory (ES) แอนติเจนได้โมโนโคลนัลแอนติบอดีต่อแอนติเจนจากผิวพยาธิ ที่ 58, 54 kD และจากลำไส้ที่ 27, 26 kD แล้วทำการ ตรวจหาแหล่งสร้างแอนติเจนซึ่งปรากฎว่าแอนติเจนที่ 58 และ 54 kD เป็น membrane antigens ที่มีอยู่มากใน เยื่อหุ้มชั้นผิวและมีอยู่บ้างในเยื่อหุ้มบางส่วนของเซลล์บุผิว ลำไส้และเซลล์ร่างกายทั่วไป ส่วนแอนติเจนที่ 27 และ 26 kD เป็นแอนติเจนในช่องของลำไส้และในไซโตปลาสซึม ของเซลล์บุผิวลำไส้ซึ่งอาจเป็นเอนไซม์สำหรับย่อยอาหาร
