Abstract
The oviducts of fully mature rice-field frogs, Rana tigerina were investigated by light (LM) and scanning electron microscopy (SEM). Special stainings using toluidine blue (TB) and alcian blue (AB) were also performed in this study. The oviduct can be divided into five parts based on the size and internal morphology. The first part is small straight tubes named pars recta (PR) . The second to the fifth part are larger coiled tubes called pars convoluta 1-4 (PC1, PC2, PC3 and PC4), respectively. Histologically, oviduct consists of three layers: mucosa, muscular and serosa. The mucosa of PR is composed of branching longitudinal mucosal folds. Epithelium consists of two cell types: ciliated cell (CC) and nonciliated cell (NCC). Non-ciliated cells contain numerous microvilli. The longitudinal rows of microvilli and cilia are alternately arranged. Muscular layer is thin and scattered under lamina propria. All parts of pars convoluta are characterized by the appearance of longitudinal or spiral epithelial ridges capped the simple tubular mucosal or mucosal glands (MGs). Epithelial ridge is comprised of CC and NCC. The openings of MG are between the ridges. With special stains, NCCs of PC1, PC3, PC4, mucosal glands of PC1 and upper mucosal glands of PC4 show metachromasia with TB and also positive staining to AB. This suggests that PCs may secrete acid mucopolysaccharide substances. The values of percentage of oviduct somatic index (%OSI) increase during breeding season from March to September; and they are 2.9, 4.7, 3.3, 3.8, 4.5, 7.3 and 3.3 respectively. This correlates with percentage of gonadosomatic index (%GSI) which also during March to September, which are 9.1, 15.5, 14, 18.7, 17.2, 17.9 and 6.6 respectively. During development, the primordial oviduct could be observed in one-month-old frogs. In two-month-old frogs, oviduct shows the appearance of hollow-tube organ. In the five-month-old frogs the oviduct can be distinguished as two main parts, PR and PC . Mucosal tubular gland can be observed from the oviducts of frogs aged five months onward. In seven-month-old frogs the oviduct can be divided into 5 parts based on external feature and histology. Oviducts in frogs aged ten months onward seem to begin to have physiological function as observed in adult frogs.
การศึกษาโครงสร้างของท่อนำไข่กบนาวัยเจริญพันธุ์ ด้วยกรรมวิธีทางจุลทรรศน์ธรรมดาและจุลทรรศน์อิเล็กตรอน แบบส่องกราด รวมทั้งย้อมด้วยสีพิเศษคือ toluidine blue และ alcian blue ท่อนำไข่กบนาสามารถแบ่งออกเป็น 5 5 ส่วนโดยพิจารณาจากขนาด ลักษณะภายนอกและโครงสร้าง ภายใน ส่วนแรกเป็นท่อตรงขนาดเล็กเรียก pars recta (PR) สำหรับส่วนที่สองถึงส่วนที่หก มีลักษณะเป็นท่อขนาดใหญ่ ไม่เท่ากันขดไปมา เรียก pars convoluta (PC) ได้แก่ PC1, PC2, PC3 และ PC4 ตามลำดับ pars recta ประกอบ ด้วยชั้น mucosa ชั้นกล้ามเนื้อ และชั้น serosa ชั้น mucosa มีการยกตัวเข้าไปในท่อนำไข่ เซลล์บุท่อส่วน PR ประกอบด้วย ciliated cell (CC) และ non-ciliated cell (NCC) NCC ประกอบด้วย microvilli เป็นจำนวนมาก cilia ของ CC และ microvilli ของ NCC มีการจัดเรียงตัวเป็นแถวยาว สลับกัน ส่วนชั้นกล้ามเนื้อบางและกระจายตัวอยู่ใต้ชั้น mucosa PC มีลักษณะเด่นชัดคือมีสันนูนเตี้ยยาวไปตามความ ยาวท่อนำไข่ สันนี้เกิดจากเยื่อบุผิวที่โค้งนูนและคลุมอยู่ ด้านบนของ mucosal glands (MG) บนสันนูนประกอบด้วย เซลล์เยื่อบุผิว 2 ชนิด คือ CC และ NCC mucosal gland มีรูเปิดอยู่ระหว่างสันนูน NCC ใน PC1 PC 3 PC4 และ MG ของ PC1 และ MG ส่วนบนของ PC4 แสดงทั้ง metachromasia ต่อสี toluidine blue และติดสี alcian blue แสดงว่าส่วน PC น่าจะมีการสร้างและหลั่งสารพวก acid mucopolysaccharide จากการศึกษา percentage of oviduct somatic index (%OSI) พบว่ามีค่าเพิ่มขึ้นในฤดูผสมพันธุ์ คือระหว่างเดือน มีนาคมถึงกันยายน โดยมีค่าเท่ากับ 2.9, 4.7, 3.3, 3.8, 4.5, 7.5 และ 3.3 ตามลำดับ ซึ่งสัมพันธ์กับ percentage of gonadosomatic index (%GSI) ที่เพิ่มขึ้นในช่วงเดียวกัน คือมีค่าเท่ากับ 9.1, 15.5, 14, 18.7, 17.2, 17.9 และ 6.6 ตามลำดับ สำหรับการพัฒนาของท่อนำไข่ของกบอายุ 1 เดือน พบว่ากลุ่มเซลล์เริ่มแรกที่จะมีการปรับสภาพไป เป็นท่อนำไข่อยู่ใกล้กับไต เซลล์กลุ่มนี้ปรับสภาพไปเป็น ท่อกลวงในกบอายุ 2 เดือน ในกบอายุ 3 เดือน สามารถแบ่ง ท่อนำไข่ออกเป็น 2 ส่วน คือ pars recta และ pars convoluta MG เห็นเป็นรูปร่างได้ชัดในกบอายุ 5 เดือน ขึ้นไป ท่อนำไข่สามารถแบ่งได้เป็น 5 ส่วนเหมือนในกบ วัยเจริญพันธุ์ได้ตั้งแต่ อายุ 7 เดือนขึ้นไป ท่อนำไข่ ของกบอายุ 10 เดือนขึ้นไป มีลักษณะโครงสร้างและขนาด คล้ายกับท่อนำไข่ของกบโตเต็มวัย
