Abstract
The cyanobaeteria are organisms those are capable of oxygenie photosynthesis. They have become an increasing popular model system for many fields of investigation via genetic and molecular biological techniques. Many attempts to manipulate the mosquito- larvicidal genes of both Bacillus thuringiensis subsp. israelensis (Bti) and B. sphearicus to be expressed in these group of organisms have proven unsatisfactory. Since all previous studies were done on the reference or laboratory strains, it is more promissing to use the strain(s) which inhabit the same place with mosquito larvae or to use the locally isolated cyanobacteria. 151 isolates of cyanobacteria were obtained from 176 water samples collected from various places in Bangkok and nearby provinces. Five isolates were sent for identification at the Pasteur Institut, France, and found to be Synechocystis PCC9001 and PCC9002 and Synechococcus PCC9003, PCC9004 and PCC9005. Due to the complication and time-consumption of conventional methods to identify cyanobacteria, identification of bacteria by DNA fingerprinting was applied to differentiate some of those local isolates. DNA from 40 isolates and 3 reference strains from the Pasteur Culture Collection (PCC) were digested with PstI and probed with (32)P-labelled Ml3 bacteriophage DNA to produce characteristic DNA fingerprinting patterns (DFP). 37 DFP were obtained. None of the local isolates gave a similar pattern to those of the reference strains. Six isolates collected from different places gave 3 pairs of identical DFP, and three isolates collected from the same place also gave identical DFP, suggesting that the ones those having identical DFP belonged to the same isolate. 31 isolates gave different patterns suggesting that cyanobacteria were highly heterogeneous. To introduce the cryIVB gene of Bti into local cyanobacteria, a shuttle vector was constructed. However transformation by natural uptake or electroporation were not successful. Using the technique of plasmid mobilization, the broad-host-range plasmid, pKT210, was able to mobilize from E.coli (habouring the helper plasmid, pRK2013) into 3 out of ll isolates of local cyanobacteria. pKT210.388, containing the cryIVB gene was mobilized into PCC9002 and CNRS2. The existence of the plasmid in cyanobacteria was demonstrated by Southern blot hybridization and rescue of plasmid in E.coli. The 128 kDa protein product from these cyanobacterial transconjugants were demonstrated by immunoblotting assays, although the amounts of products were quite low. However, the results indicated that the cryIVB gene of Bti was able to be introduced and expressed in locally isolated cyanobacteria. In an attempt to improve the expression of cryIVB gene in local cyanobacteria, pKT210.388.T7.pol which contained T7 promoter upstream to the cryIVB gene, T7 RNA polymerase gene and the whole set of its regulatory genes was constructed. In E.coli the cryIVB gene in this plasmid was shown to be expressed very well after heat induction. The cells were also highly toxic to mosquito larvae. However, expression in cyanobacteria using the same system was not higher than the expression under its own promoter . It was demonstrated by rescue of plasmids in E.coli that the cryIVB gene in plasmids from cyanobacterial had not been changed within the cyanobacterial cells as the gene was able to express in the rescued E.coli to a similar extent as in cells that harboring the original plasmid . The low expression might be due to the low copy number of the broad host range plasmid, pKT210, in cyanobacteria.
ปัจจุบันได้มีผู้นิยมใช้ cyanobacteria ซึ่งเป็น บักเตรีสังเคราะห์แสงที่พบได้ในแหล่งน้ำทั่วไปมาใช้ใน การศึกษาทางด้านอณูชีววิทยากันอย่างกว้างขวาง ในวิทยานิพนธ์นี้ต้องการที่จะศึกษาการใช้บักเตรีชนิดนี้ ในการเป็น host เพื่อผลิตสารพิษฆ่าลูกน้ำยุงจาก gene cryIVB ของ Bacillus thuringiensis subsp. israelensis (Bti) เพื่อเป็นแนวทางในการนำไปสู่ การแก้ปัญหาของการใช้ Bti ในการควบคุมปริมาณของ ลูกน้ำยุงในธรรมชาติ จาก 176 ตัวอย่างน้ำที่เก็บมาจากแหล่งน้ำต่าง ๆ ในกรุงเทพมหานคร และเขตปริมณฑล พบเชื้อ cyanobacteria 151 isolates, ในจำนวนนี้ 5 isolates ได้รับการ identify เพื่อแยกชนิดจากสถาบัน Pasteur Institut ในประเทศฝรั่งเศส ให้เป็น synechocystis PCC9001, PCC9002 และ Synechococcus PCC9003, PCC9004 และ PCC9005 ในการศึกษาได้นำวิธี M13-DNA fingerprinting technique มาใช้ในการจำแนกชนิดของ local strains ของ cyanobacteria จำนวน 40 isolates เทียบกับ 3 strains ของ reference strains โดยการตัด DNA ของเชื้อเหล่านี้ ด้วยเอ็นไซม์ตัดจำเพาะ Pst และตรวจด้วยตัวติดตาม (32)P- labelled M13 probe พบว่า DNA fingerprinting patterns (DFP) ที่ได้ สามารถถูกจัดแบ่งออกได้เป็น 37 แบบ, ไม่มี local strains ตัวใดให้ DFP ที่เหมือนกับ DFP ของ reference strains และเมื่อเปรียบเทียบระหว่าง local strains ด้วยกันเอง พบว่า เชื้อบางตัวที่ได้มาจากแหล่งน้ำ ที่ต่างกันให้ DFP ที่เหมือนกัน แสดงว่าเชื้อที่ให้ DFP ที่เหมือนกัน น่าจะเป็น isolates เดียวกัน ผลการทดลองสามารถสรุปได้ว่า วิธีของ M13-DNA fingerprinting สามารถนำมาใช้ในการจำแนก ชนิดของเชื้อ cyanobacteria ได้ ในการพยายามที่จะนำ crylVB gene เข้าสู่ local strains ของ cyanobacteria โดยการสร้าง shuttle vectors จาก endogenous plasmids ปรากฎว่า ไม่สามารถจะนำ crylVB gene เข้าสู่ cyanobacteria ที่ทดลองได้ โดยวิธีของ plasmid mobilization, สามารถทำการ mobilize plasmid pKT210 (broad-host-range plasmid) เข้าสู่ local strains ของ cyanobacteria PCC9003, PCC9004 และ CNRS2 ได้, จึงได้ทำการตัดต่อ gene crylVB เข้ากับ pKT210 plasmid ได้ plasmid ที่ชื่อว่า pKT210.388 และ ทำการ mobilize เข้าสู่ PCC9003 และ CNRS2 จากการตรวจสอบ โดย Southern blot hybridization และ rescue เข้าสู่ E.coli พบว่ามี plasmid pKT210.388 อยู่ใน cyanobacteria 2 isolates นั้นจริง และยังสามารถตรวจพบโปรตีนขนาด 128 kDa จากเชื้อทั้ง 2 ได้ แต่ปริมาณที่ได้ค่อนข้างต่ำ, จึงได้พยายาม ปรับปรุงการสร้างโปรตีนขนาด 128 kDa จาก crylVB gene นี้ โดยการสร้าง plasmid pKT210.388.T7.pol ซึ่งประกอบด้วย T7 promoter อยู่ upstream จาก cryIVB และ T7 RNA polymerase gene รวมทั้ง regulatory genes ทั้งชุด ใน E.coli พบว่าหลังจาก heat induction มีการสร้างโปรตีนขนาด 128 kDa จาก cryIVB gene ใน plasmid ตัวใหม่นี้ในปริมาณที่สูงมาก และ cells พวกนี้สามารถฆ่าลูกน้ำยุงได้เป็นอย่างดี อย่างไรก็ตาม, ใน cyanobacteria การสร้างโปรตีนขนาด 128 kDa จาก cryIVB gene จาก plasmid เดียวกันนี้ไม่สามารถถูกกระตุ้นให้เพิ่ม ปริมาณได้โดย heat induction แม้ว่าสามารถตรวจสอบได้ว่า มี pKT210.388.T7.pol อยู่ใน cyanobacteria จริง การที่ ตรวจพบโปรตีนขนาด 128 kDa ในปริมาณที่ค่อนข้างต่ำนั้น อาจจะเนื่องมาจากจำนวน (copy number) ของ pKT210 plasmid ใน cyanobacteria มีน้อยมาก ทำให้ผลผลิตของ cryIVB gene ที่ได้น้อยไปด้วย อย่างไรก็ตามการศึกษานี้สามารถแสดงให้เห็นถึงความ สำเร็จในการนำ cryIVB gene เข้าสู่ local strain ของ cyanobacteria รวมถึงความสามารถในการสร้างโปรตีนขนาด 128 kDa ใน cyanobacteria นั้น ๆ ด้วย
