Abstract
การศึกษาในครั้งนี้มีวัตถุประสงค์เพื่อหาค่าความไวและความจาเพาะของชุดตรวจ Pall eBDS, pH meter และ urine dip strip ซึ่งการศึกษาในครั้งนี้ : นำ LPPC 1 ถุง มาแบ่งเป็น 6 ถุง ถุงละ 55 ml(LPPC ที่ใช้ทั้งหมด 15 ถุง โดยแต่ละถุงนำมาแบ่งเป็น 6 ถุงเล็ก ทาให้ได้ตัวอย่างทั้งหมด 90 ถุง สำหรับฉีดเชื้อมาตรฐานเชื้อละ 15 ถุง และ negative control 15 ถุง) จากนั้นฉีดเชื้อแบคทีเรียมาตรฐาน 5 เชื้อ ได้แก่ Escherichia coli ( ATCC 25922 ), Staphylococcus epidermidis (ATCC 49134), Staphylococcusaureus ( ATCC 27217 ), Bacillus cereus ( ATCC 7064 ), Pseudomonas aeruginosa(ATCC 27853 ) โดยใช้ความเข้มข้นของเชื้อแบคทีเรีย 60 CFU/ml โดยฉีดถุงละ 1 เชื้อ ส่วนถุงที่ 6 ไม่ต้องฉีดเชื้อ เก็บไว้ทำ negative control หลังจากฉีดเชื้อเสร็จแล้วเก็บไว้ที่ตู้เก็บเกล็ดเลือด ประมาณ 30 นาที จึงแบ่งเกล็ดเลือด 3 ml ใส่ชุดตรวจ Pall eBDS อ่านผล 24 ชั่วโมงหลังจากเก็บที่ตู้เก็บเกล็ดเลือด ส่วนถุงเกล็ดเลือดนั้นให้ตัดสายปล้องถุงเลือดเพื่อเพาะเชื้อด้วย blood agar พร้อมทั้งวัดค่า pH ด้วย pH meter และ Urine dipstick ทุกวัน จนกว่าจะได้ผลบวกหรือจนกว่า เกล็ดเลือดจะหมดอายุ จากผลการศึกษาพบว่า ชุดตรวจ Pall eBDS ให้ ผลบวกกับเกล็ดเลือดที่ฉีดเชื้อทั้ง 75 ถุง และให้ผลลบกับเกล็ดเลือดที่ไม่ได้ฉีดเชื้อทั้ง 15 ถุง (negative control ในขณะที่ pH meter และ urine dip strip ให้ผลบวกหลังฉีดเชื้อ 72 ชั่วโมง ทั้ง 75 ถุง จากผลการศึกษาสรุปได้ว่า ชุดตรวจ Pall eBDS มี sensitivity และ specificity 100% ต่อเชื้อ Escherichia coli, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus cereus, Pseudomonas aeruginosa ในกรณีที่เชื้อปนเปื้อน 60 CFU/ml ขึ้นไป ในขณะที่ pH meter และ urine dipstick สามารถตรวจพบเชื้อได้เมื่อเชื้อ มีปริมาณมากกว่า 1x1010 CFU/ml (Escherichia coli, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus cereus, Pseudomonas aeruginosa)
