Abstract
A lipase-producing microorganism yeast strain was isolated from raw milk. It was identified as Trichosporon asteroides LP005. Optimum physical condition for cultivation was at 30 degree C temperature with agitation 200 rpm. A production medium has been developed for T. asteroides LP005 lipase which composes of mineral salts of medium A supplemented with 0.5% soybean meal, 0.5% beef extract and 0.5% palm oil. The purification process of T. asteroides LP005 lipase included ammonium sulphate precipitation at 70% saturation and gel filtration chromatography on Sephadex G-200. The molecular weight of the lipase determined by SDS-PAGE was 45 kDa. The optimum pH and temperature of T. asteriodes LP005 for activity are pH 5.0 and 60 degree C, respectively. This lipase was stable at acidic condition and temperature lower than 70 degree C. The chelating agent, EDTA, did not affect the activity of the enzyme which suggesting that it was not a metalloenyme. T. asteroides LP005 lipase was positional non-specificity. The ability of this lipase to efficiently hydrolyze tuna oil indicated its potential to be applied for the production of EPA and DHA. T. asteroides LP005 could increase the concentration of EPA and DHA approximately 33% and 47%, respectively. T. asteroides LP005 lipase could also potentially be used for lipid-riched wastewater treatment as it could decrease the BOD value of the wastewater from S&P bakery factory by approximately 50%.
