This study aimed to assess the efficacy of fluoride toothpaste containing 1500 ppm fluoride with and without sodium lauryl sulfate (SLS) in preventing root caries in a human root caries model by evaluating fluoride ion disintegration, inhibition of Streptococcus mutans adhesion, and demineralization inhibition. Laboratory experiments involved preparing toothpaste solutions with and without SLS for comparison. Fluoride dissociation was measured using a fluoride ion selective electrode, Streptococcus mutans adhesion inhibition was assessed using a 96-well plate biofilm formation assay, and demineralization properties were compared. Thirty healthy human root dentin samples were divided into five groups and cultured with Streptococcus mutans in a human root caries model for 10 days. There were no statistically significant differences in fluoride dissociation efficiency, inhibition of Streptococcus mutans adhesion, or demineralization inhibition between toothpaste with and without SLS in the human root caries model. Both fluoride toothpastes containing 1500 ppm fluoride with and without SLS showed comparable efficacy in fluoride ion dissociation, demineralization inhibition, and prevention of Streptococcus mutans adhesion.
