Evaluation of partially purified cholesterol esterase for clinical assay of cholesterol
(Mahidol University. Mahidol University Library and Knowledge Center, 1989) Koson Sim; Pichit Tosukhowong
Bhinyo Panijpan ths."Serum cholesterol determination is a routine diagnostic method for arteriosclerosis. The enzymatic determination of cholesterol requires three enzymes, namely, cholesterol esterase, cholesterol oxidase and peroxidase. The objective of this project was to investigate a new source of cholesterol esterase suitable for diagnostic application. Forty five isolates of bacteria and sixteen isolates of moulds were screened for extracellular cholesterol esterase. The isolate that produced the highest yield of the enzyme was selected and was identified as Pseudomonas sp. based on Bergey's manual. The enzyme was partially purified by 2 methods: one method by the combination of DEAE-cellulose column and Sephadex G-200 gel column chromatography, and the other method by calcium chloride precipitation. The purification fold of the enzyme were about 24 and 13 respectively. The specific activity of the partially purified enzyme was about 1.2 unit/mg protein with 75% recovery for both methods of purification. The partially purified enzyme was studied for its physical and biochemical properties, and the results were the followings: molecular weight was about 600,000 daltons by gel filtration, pH optimum 7.0-9.0, temperature optimum 50 degree C, pH stability in the range 5.0-10.0, K(,m) value 3.3x10 (-5) M, stable at 4 degree C and in frozen condition for at least 2 months, the enzyme was able to hydrolyze serum cholesterol esters to the same extent as alkaline hydrolysis. These results suggest that cholesterol esterase from this pseudomonad was potentially suitable for further development in diagnostic application.

Evaluation of the protective effects of atropine and diazepam against cypermethrin induced lethality
(Mahidol University. Mahidol University Library and Knowledge Center, 2024) Punthip Teeyapant; Jutamaad Satayavivad; Udom Chantharaksri ; Yupin Sanvarinda
The protective effects of atropine and diazepam against cypermethrin induced lethality were evaluated in mice. Diazepam was used clinically to control convulsion induced by toxic dose of cypermethrin. It was found that diazepam delayed the onset

Pseudomonas pseudomallei : immunological studies with special application for the diagnosis of melioidosis
(Mahidol University. Mahidol University Library and Knowledge Center, 2024) Huttaya Wuttipongwaragid; Stitaya Sirisinha; Surasakdi Wongratanacheewin; Sansanee Chaiyaroj
Melioidosis, an infection caused by Pseudomonas (Ps.) pseudomallei , is considered to be one of the important health problems in Thailand. Most of the patients reside in the northeastern part of the country. The clinical features may vary from... asymptomatic to acute and fatal septicemia, with a mortality rate of 80-90% in untreated cases. The disease cannot be readily diagnosed by clinical pictures because its signs and symptoms mimic a number of other infectious diseases. Consequently, a definitive

Detection and identification of dengue viruses in suckling mice : comparison studies between the Western blot and viral gene amplification processes
(Mahidol University. Mahidol University Library and Knowledge Center, 1992) Chalenee Watanawanna; Vina Churdboonchart; Natth Bhamarapravati; Sakol Panyim
of tissue, several methods to extract viral RNA from these tissues were employed and the results were evaluated. The results indicated that the PCR method can detect viral genes in brain, liver, spleen, lymph nodes and which confirmed previous study using... VAS/EIA. Improvement can be done via better extraction method which may lead to a sensitive system to detect dengue viral RNA in plasma and serum of infected mouse with an ultimate goal to identify the virus in clinically confirmed dengue fever cases.

Identification of specific antigen of Pseudomonas pseudomallei and monoclonal antibody production for the diagnosis of melioidosis

for the detection of serum antibodies. In this study, fractionated 19.5-kDa antigen was prepared from unfractionated whole cell antigen of P. pseudomallei and evaluated for its potential usefulness in the diagnosis of acute septicemic melioidosis. The antigen... it to be promising for detecting antigen in the urine. However, whether or not this method will be useful in the diagnosis of acute melioidosis depends on further investigation when clinical specimens are available for analysis.

Production and characterization of monoclonal antibodies against protein antigens of Pseudomonas pseudomallei
(Mahidol University. Mahidol University Library and Knowledge Center, 1991) Ratchanee Atthi; Stitaya Sirisinha; Pranee Sithisarn; Surasakdi Wongratanacheewin
Melioidosis is a disease caused by Pseudomonas pseudomallei. In Thailand, most of the patients lived in the northeast. At present, melioidosis oases were increasingly reported. The clinical manifestations are variable tanging from chronic to aoute... and was evaluated for its potential in detecting the Ps. pseudomallei antigen. It was found that this clone could detect the protein antigen of Ps. pseudomallei in aqueous solution at a concentration as low as 15 ng/ml by B-SA ELISA. However , this clone failed

Detection for dengue virus infection using suckling mice as a biological amplification system and enzymeimmunoassay
(Mahidol University. Mahidol University Library and Knowledge Center, 2024) Marisa Kemapunmanus; Vina Churdboonchart; Natth Bhamarapravati; Sakol Punyim
Detection for dengue viral proteins in patients sera with dengue virus infection was investigated by using suckling mouse as a biological amplification system evaluated in conjunction with the Viral antigens strip/ enzymeim munoassay (VAS/EIA.... Two groups of patients sera with clinical diagnosis of dengue infection collected from Ramathibodi hospital were tested for this study. Three dilutions, 1:1, 1:3 and 1:10 of patients sera were compared to select the proper serum dilution

Prevalence of asymptomatic genital herpes in near-term pregnant women and anti-HSV-2 IgM response following first episode and recurrent diseases
(Mahidol University. Mahidol University Library and Knowledge Center, 1988) Wanna Pumeechockchai; Chalobon Yoosook; Pantipa Sinarachatanant
shedding shold be done during the last month of gestation in women with a history of genital herpes. In the present study, the prevalence of asymptomatic genital herpes in near-term pregnant (at least 30 weeks of gestation) who attended the antenatal clinic... of immunoperoxidase staining of tissue smears cannot be evaluated since specimens from pregnant women who excreted the virus are unavailable. Staphylococcal co-agglutination test could not detect HSV antigens in all 3 HSV-isolation and B-SA ELISA positive samples
